MUELLER-HINTON AGAR ACCORDING TO NCCLS - Introduction

MH(A) medium is a commonly used microbial culture medium. Its properties, uses, preparation methods and safety information are as follows:

Nature:
MH(A) medium is a colorless to pale yellow transparent liquid composed of a variety of organic substances, carbohydrates, amino acids, vitamins and minerals. It usually has a pH between 7.2 and 7.4 and contains 0.2 percent peptone.

Use:
MH(A) medium is mainly used for the growth and reproduction of microorganisms. It is suitable for a variety of bacteria, fungi and yeast culture, can be used for regular colony count, bacterial identification and drug sensitivity test and other experimental research.

Preparation Method:
The method of preparing MH(A) medium is as follows:
1. Dissolve 15 grams of broth powder (or other sources equivalent to broth powder) in an appropriate amount of distilled water;
2. Add 0.2 grams of peptone and fully stir and dissolve;
3. make up to 1000 ml with distilled water and adjust the pH value to 7.2-7.4;
4. without sterilization, can be used directly.

Safety Information:
MH(A) medium is relatively safe. But still need to pay attention to the following points:
1. use should pay attention to personal protection, wear experimental gloves and other protective measures;
2. avoid swallowing, inhalation or contact with eyes or skin, such as accidental contact, should immediately rinse with plenty of water;
3. storage should avoid high temperature, direct sunlight and humid environment.
